If you have a Discount Coupon, it can be redeemed at the checkout during its period of validity. Simply enter the code in the 'redemption code' box during the checkout process.
Gift Certificates are different. If you redeem a Gift Certificate, the amout will be credited to your account and you can use as much or as little against each purchase. They can also be forwarded to friends.
